β-Carotene is a naturally occurring, bioactive compound found in various fruits and vegetables. Determining its concentration in foods is of great nutritional importance; however, it is challenging due to matrix interference. In this study, A simple, rapid, and sensitive method of homogeneous liquid-liquid microextraction using a switchable hydrophilicity solvent (SHS-HLLME) was optimized for the extraction and determination of β-carotene in complex food matrices (sweet potato and cassava genotypes) prior to HPLC-DAD analysis. The optimal extraction conditions consisted of using N,N-dimethylcyclohexylamine (DMCHA) as the switchable hydrophilicity solvent (SHS), 1000 μL of a mixture of SHS: 6 mol L⁻¹ HCl (1:1 v/v), 600 μL of 10 mol L⁻¹ NaOH, and 1000 μL of 2.0% (w/v) NaCl. Under the optimized conditions, the proposed method exhibited a linear range of 1–40 µg L⁻¹, with limits of detection and quantification of 0.305 µg L⁻¹ and 0.926 µg L⁻¹, respectively. The intra-day and inter-day precision of the developed method yielded RSD values in the range of 1.1–3.7% and 1.2–3.6%, respectively. The recovery rates for β-carotene extraction ranged from 97.9% to 107.0%, with RSD values less than 5%. The determination of β-carotene in real samples was performed in just 3.5 minutes using the proposed methodology. The results demonstrated that the SHS-HLLME approach is fast, simple, eco-friendly, and provides satisfactory analytical performance.
